Functionele huidcellen
De buitenste laag van de huid wordt de opperhuid genoemd. Deze bestaat uit verschillende lagen en bevat vier belangrijke celtypen
  • Keratinocyten (barrièrecellen),
  • Melanocyten (pigmentcellen),
  • Langerhanscellen (immuuncellen) en
  • Merkelcellen (zintuigcellen).
Functional Skin Cells
Huidcellen worden bij elkaar gehouden door desmosomen en de cellen worden beschermd en verstevigd door keratohyaline en lamellaire granules.

Keratinocytes, Melanocytes & Melanin

Keratinocyten vormen ~ 90% van de celpopulatie. cellen. Keratinocyten ontwikkelen zich vanuit de onderste of basale laag van de epidermis en migreren dan omhoog over een periode van ongeveer vier weken naar het buitenoppervlak (stratum corneum) waar ze worden afgestoten. Zowel keratinocyten en melanocyten produceren belangrijke eiwitten die de groei en vermenigvuldiging van melanocyten en de productie van melanine regelen.
De melanocyt is een zeer gespecialiseerde, bijzonder lang levende cel die melanine (natuurlijk huidpigment/kleurstof) produceert uit het aminozuur tyrosine. pigment/kleur) produceert uit het aminozuur tyrosine. Binnen de onderste laag van van de opperhuid zijn er ongeveer 10x meer keratinocyten dan melanocyten; Er zijn echter nog steeds ongeveer 1200 melanocyten per mm2 huid, onafhankelijk van ras. De dichtheid van melanocyten in de huid hangt voornamelijk af van blootstelling aan ultraviolette straling en factoren die worden afgescheiden door keratinocyten en fibroblasten (functionele cellen in de dermis, de huidlaag onder de opperhuid).
Melanine gemaakt in de melanocyt wordt overgedragen aan basale keratinocyten, waarbij één melanocyt tot wel 36 keratinocyten. Melanine heeft veel eigenschappen die gunstig zijn voor het lichaam, zoals lichaam, waaronder absorptie en verstrooiing van UV-licht, vrije radicalen vangen, redoxbalans en ionenopslag. De melanocyt scheidt ook verschillende signaalmoleculen die niet alleen gericht zijn op keratinocyten, maar ook op belangrijke huidcellen van het immuunsysteem.

Keratinocyten en melanocyten zijn noodzakelijk voor een normale, gezonde huid functie en kleur. Thymoquinone is widely regarded as being the biologically active component of Nigella Sativa.

Thymoquinone is likely to have a dual action in the remediation of vitiligo

  • It can trigger a multi-step, biochemical pathway, that has a stimulatory effect on melanin production, as has been demonstrated in animals.
A recent human-clinical trial(27) conducted in 2019 demonstrated that a cream containing Nigella sativa resulted in statically significant re-pigmentation in the hands, face and genital region of 47 men and women with vitiligo after 6 months of use, and was considered to be especially suitable for use on delicate skin.
An earlier double-blind randomised human clinical trial(13) compared the effects of Nigella sativa and Fish Oil in resolving the symptoms of Vitiligo. Nigella oil was found to be more effective, with the Vitiligo Area Scoring Index reducing from 4.98 to 3.75, with pronounced improvements in the upper extremities, trunk, head and neck after 6 months of use.
As is the case of Psoralen and Berberine; Thymoquinone utilises different inter and intra-cellular processes and different biochemistry in effecting its antioxidant and stimulatory effects.
